Refreshing registry key programatically

i have created a application in which i will be writing a registry key to users node so that application will use in the later execution. Is there a way that registry keys can be refreshed programatically using vb.net with out closing and opening the registry keys again

What I have tried:

not sure how to proceed further

Why would you have to shutdown the application to update a registry key in the first place? What is your current code handling this requirement, and what happens when it runs that makes you say it is incorrect?
