Is there any code or possibilities to find if there is a screen reader on my PHP site.

What I have tried:

Currently we have no idea on how to handle this for 508 Compliance.

I don't understand your question.

No, I don't think there is any way for your code to know that a screen reader is reading it to someone. Why would you care?

You cannot detect such, but you could provide the basics...alt text for images and videos, relative fonts so that users with impaired vision (but not blind) can easily scale etc

There are tools to check you existing site. See also What is 508 Compliance? | 508Checker.com[508 Compliance] where you can scan your site to check for compliance.
