can we do some changes in master page when content page already added in master page?

Yes, you can change the master page any way you like, so long as you don't remove the content areas your content pages are referencing. That's kind of the point, you have one page you can update, and it update all the pages that use it.

Hi, you can change the Master Page content, but after changing it in order to see the changes in the Content Page, you have to refresh the Content Page.

Also do not remove the Content Place Holder from the Master Page which are being referred in the Content Page.
