Large directory on IIS

I have a shared folder on the server with an approximate size of 54 GB. I want to make it accessible from a web application by mapping it to a virtual directory on IIS.
My question is:
Does it make any problem with the IIS if I create that virtual directory?

No. It is - as you already stated - a virtual directory and for that it is just like a link on your desktop...
No overwhelm whatsoever to IIS...
